Married Isabella of Capua (1510 – 1559) daughter of Ferrante of Capua, Count of Alessano, Marquess of Specchia, Duke of Termoli and Prince of Molfetta.

• Ferrante III Gonzaga (1618 – 1678) Duke of Guastalla, Prince of Molfetta. Knight of the Order of Santiago. Married Margherita (1619 – 1692) daughter of Alfonso III d'Este, Duke of Modena. He died without male heirs, the Duchy of Guastalla became officially a part of the Duchy of Milan ruled by Charles IV of Gonzaga-Nevers (1652 – 1708) Duke of Mantua and Montferrat 1678-1693, married to Anna Isabella Gonzaga (1655 – 1703) elder daughter of Ferrante III Gonzaga, Duke of Guastalla.

• Giuseppe Gonzaga (1690 – 1746) Duke of Guastalla. Married Eleonore von Holstein (1715–1760) daughter of Duke Leopold of Schleswig-Holstein-Sonderburg-Wiesenburg.

In 1748 at the Treaty of Aix-La-Chapelle, Guastalla was ceded to the Spanish, together with the Duchy of Parma and Piacenza, with which it was merged.
